The present study aimed to estimate the height growth curve for Mexican boys and girls based on their body mass index (BMI) status (normal and overweight/obese) and to develop a height Lambda, Mu, and Sigma (LMS) growth reference for Mexican children aged 2 to 18 years. Methods: Chronological age and height records (7,097 boys and 6,167 girls) were obtained from the Mexican National Survey of Health and Nutrition database. Height growth curves were fitted using the Preece-Baines 1 (PB1) model and the LMS method. Results: Age at peak height velocity (APHV) was 12.4 and 12.7 years for overweight-obese and normal-weight boys, respectively, and was 9.6 and 10.4 years for overweight-obese and normal-weight girls, respectively. Growth velocity was higher at the age of take-off (TO) in overweight-obese children than in normal-weight children (5.2 cm/year vs. 5 cm/year in boys and 6.1 cm/year vs. 5.6 cm/year in girls); nevertheless, the growth velocity at APHV was higher for normal-weight children than for overweight-obese children (7.4 cm/year vs. 6.6 cm/year in boys and 6.8 cm/year vs. 6.6 cm/year in girls, respectively). Distance curves developed in the present study and by the World Health Organization (WHO) using LMS showed similar values for L and S parameters and a higher M value compared with the WHO reference values. Conclusion: This study concluded that overweight-obese children had earlier APHV and lower PHV than normal-weight children. Furthermore, Mexican children and adolescents were shorter than the WHO growth reference by age and sex.

To efficiently detect the maturity stages of Camellia oleifera fruits, this study proposed a non-invasive method based on hyperspectral imaging technology. First, a portable hyperspectral imager was used for the in-field image acquisition of Camellia oleifera fruits at three maturity stages, and ten quality indexes were measured as reference standards. Then, factor analysis was performed to obtain the comprehensive maturity index (CMI) by analyzing the change trends and correlations of different indexes. To reduce the high dimensionality of spectral data, the successive projection algorithm (SPA) was employed to select effective feature wavelengths. The prediction models for CMI, including partial least squares regression (PLSR), support vector regression (SVR), extreme learning machine (ELM), and convolutional neural network regression (CNNR), were constructed based on full spectra and feature wavelengths; for CNNR, only the raw spectra were used as input. The SPA-CNNR model exhibited more promising performance (RP = 0.839, RMSEP = 0.261, and RPD = 1.849). Furthermore, PLS-DA models for maturity discrimination of Camellia oleifera fruits were developed using full wavelength, characteristic wavelengths and their fusion CMI, respectively. The PLS-DA model using the fused dataset achieved the highest maturity classification accuracy, with the best simplified model achieving 88.6 % accuracy in prediction set. This study indicated that a portable hyperspectral imager can be used for in-field determination of the internal quality and maturity stages of Camellia oleifera fruits. It provides strong support for non-destructive quality inspection and timely harvesting of Camellia oleifera fruits in the field.

Glycoproteins located on the cell surface play a pivotal role in nearly every extracellular activity. N-glycosylation is one of the most common and important protein modifications in eukaryotic cells, and it often regulates protein folding and trafficking. Glycosylation of cell-surface proteins undergoes meticulous regulation by various enzymes in the endoplasmic reticulum (ER) and the Golgi, ensuring their proper folding and trafficking to the cell surface. However, the impacts of protein N-glycosylation, N-glycan maturity, and protein folding status on the trafficking of cell-surface glycoproteins remain to be explored. In this work, we comprehensively and site-specifically studied the trafficking of cell-surface glycoproteins in human cells. Integrating metabolic labeling, bioorthogonal chemistry, and multiplexed proteomics, we investigated 706 N-glycosylation sites on 396 cell-surface glycoproteins in monocytes, either by inhibiting protein N-glycosylation, disturbing N-glycan maturation, or perturbing protein folding in the ER. The current results reveal their distinct impacts on the trafficking of surface glycoproteins. The inhibition of protein N-glycosylation dramatically suppresses the trafficking of many cell-surface glycoproteins. The N-glycan immaturity has more substantial effects on proteins with high N-glycosylation site densities, while the perturbation of protein folding in the ER exerts a more pronounced impact on surface glycoproteins with larger sizes. Furthermore, for N-glycosylated proteins, their trafficking to the cell surface is related to the secondary structures and adjacent amino acid residues of glycosylation sites. Systematic analysis of surface glycoprotein trafficking advances our understanding of the mechanisms underlying protein secretion and surface presentation.

BACKGROUND AND AIMS: Stand-replacing crown fires are the most prevalent type of fire regime in boreal forests in North America. However, a substantial proportion of low-severity fires are found within fire perimeters. Here we aimed to investigate the effects of low-severity fires on the reproductive potential and seedling recruitment in boreal forests stands in between stand-replacing fire events. METHODS: We recorded site and tree characteristics from 149 trees within twelve sites dominated by mature black spruce [Picea mariana (Mill.) B.S.P.] trees in the Northwest Territories, Canada. The presence of fire-scarred trees supported classification of sites as unburned or affected by low-severity fires in recent history. We used non-parametric tests to evaluate differences in site conditions between unburned and low-severity sites, and mixed effect models to evaluate differences in tree age, size, and reproductive traits among unburned trees and trees from low-severity sites. KEY RESULTS: Results showed significantly higher density of dead black spruce trees in low-severity sites, and marginally significant higher presence of permafrost. Trees from low-severity fire sites were significantly older, exhibited significantly lower tree growth, and showed a tendency towards a higher probability of cone presence and percentage of open cones compared to trees from unburned sites. Surviving fire-scarred trees affected by more recent low-severity fires showed a tendency towards higher probability of cone presence and cone production. Density of black spruce seedlings significantly increased with recent low-severity fires. CONCLUSIONS: Trees in low-severity sites appeared to have escaped mortality from up to three fires, as indicated by fire scar records and their older ages. Shallow permafrost at low-severity sites may cause lower flammability, allowing areas to act as fire refugia. Low-severity surface fires temporarily enhanced the reproductive capacity of surviving trees and the density of seedlings, likely as a stress response to the fire event.

BACKGROUND: Informativeness, in the context of clinical trials, defines whether a study's results definitively answer its research questions with meaningful next steps. Many clinical trials end uninformatively. Clinical trial protocols are required to go through reviews in regulatory and ethical domains: areas that focus on specifics outside of trial design, biostatistics, and research methods. Private foundations and government funders rarely require focused scientific design reviews for these areas. There are no documented standards and processes, or even best practices, toward a capability for funders to perform scientific design reviews after their peer review process prior to a funding commitment. MAIN BODY: Considering the investment in and standardization of ethical and regulatory reviews, and the prevalence of studies never finishing or failing to provide definitive results, it may be that scientific reviews of trial designs with a focus on informativeness offer the best chance for improved outcomes and return-on-investment in clinical trials. A maturity model is a helpful tool for knowledge transfer to help grow capabilities in a new area or for those looking to perform a self-assessment in an existing area. Such a model is offered for scientific design reviews of clinical trial protocols. This maturity model includes 11 process areas and 5 maturity levels. Each of the 55 process area levels is populated with descriptions on a continuum toward an optimal state to improve trial protocols in the areas of risk of failure or uninformativeness. CONCLUSION: This tool allows for prescriptive guidance on next investments to improve attributes of post-funding reviews of trials, with a focus on informativeness. Traditional pre-funding peer review has limited capacity for trial design review, especially for detailed biostatistical and methodological review. Select non-industry funders have begun to explore or invest in post-funding review programs of grantee protocols, based on exemplars of such programs. Funders with a desire to meet fiduciary responsibilities and mission goals can use the described model to enhance efforts supporting trial participant commitment and faster cures.

PURPOSE: To examine the distribution and characteristics of pediatric foot fractures on radiographs with respect to age and skeletal maturation, and to identify predictors of surgery. METHODS: This retrospective study included children (≤ 18 years) with foot fractures, who underwent radiographic examinations (2020-2022). Electronic medical records were reviewed to obtain demographic and clinical data. Fracture characteristics, including anatomic location, presence of displacement, angulation, articular involvement, and, if skeletally immature, physeal involvement and Salter-Harris fracture pattern were collected. Logistic regression models were used to identify predictors of surgery. RESULTS: 1,090 (596-boys, 494-girls; mean age, 11.0 ± 4.0 years) patients with 1,325 (59.8% metatarsal, 33.8% phalangeal, and 6.4% tarsal) fractures were included. Fractures of 1st metatarsal were more common among younger children whereas fractures of 2nd-4th and 5th metatarsals were more common among older children (median ages: 5.9 years vs. 10.3 years and 12.4 years, p < 0.001). Intra-articular fractures were more common among maturing and mature than immature bones (25.3% and 20.4% vs. 9.9%, p < 0.001). Physeal involvement was uncommon (162/977, 16.6%) and the most common pattern was Salter-Harris type II (133/162, 82.1%). A minority (47/1090, 4.3%) of patients required surgery and independent predictors of surgery included physeal involvement (OR = 5.12, 95% CI: 2.48-10.39, p < 0.001), multiple fractures (OR = 3.85, 95% CI: 1.67-8.53, p = 0.001), fracture displacement (OR = 9.16, 95% CI:4.43-19.07, p < 0.001), and articular involvement (OR = 2.72, 95% CI:1.27-5.72, p = 0.008). Using these predictors, the likelihood for surgery ranged between 8.0% with 1 and 86.7% with 3 predictors. CONCLUSION: Pediatric foot fracture patterns differed based on age and regional skeletal maturation. Physeal involvement, multiple fractures, fracture displacement, and articular involvement were independent predictors of surgery in our study group.

Purpose: Greulich and Pyle is the most used system to estimate skeletal maturity but has significant drawbacks, prompting the development of newer skeletal maturity systems, such as the modified Fels skeletal maturity systems based on knee radiographs. To create a new skeletal maturity system, an outcome variable, termed a "skeletal maturity standard," must be selected for calibration of the system. Peak height velocity and 90% of final height are both considered reasonable skeletal maturity standards for skeletal maturity system development. We sought to answer two questions: (1) Does a skeletal maturity system developed using 90% of final height estimate skeletal age as well as it would if it was instead developed using peak height velocity? (2) Does a skeletal maturity system developed using 90% of final height perform as well in lower extremity length prediction as it would if it was instead developed using peak height velocity? Methods: The modified Fels knee skeletal maturity system was recalibrated based on 90% of final height and peak height velocity skeletal maturity standards. These models were applied to 133 serially obtained, peripubertal antero-posterior knee radiographs collected from 38 subjects. Each model was used to estimate the skeletal age of each radiograph. Skeletal age estimates were also used to predict each patient's ultimate femoral and tibial length using the White-Menelaus method. Results: The skeletal maturity system calibrated with 90% of final height produced more accurate skeletal age estimates than the same skeletal maturity system calibrated with peak height velocity (p < 0.05). The 90% of final height and peak height velocity models made similar femoral and tibial length predictions (p > 0.05). Conclusion: Using the 90% of final height skeletal maturity standard allows for simpler skeletal maturity system development than peak height velocity with potentially more accuracy.

BACKGROUND: The likelihood of healing of osteochondritis dissecans decreases with skeletal maturity and there are theories that abnormal biomechanical forces contribute to the development and progression of these lesions. OBJECTIVE: To characterize, according to regional skeletal maturity, the morphology and alignment indices of the patellofemoral joint on MRI in patients with patellar osteochondritis dissecans. MATERIALS AND METHODS: MRI examinations of patients with patellar osteochondritis dissecans obtained between January 2008 and May 2023 were retrospectively reviewed to determine regional skeletal maturity, osteochondritis dissecans lesion size and location, patellar and trochlear morphology (Wiberg/Dejour classifications), and to calculate trochlear sulcus angles, trochlear depth index, lateral trochlear inclination, Insall-Salvati index, Caton-Deschamps index, patellar tendon-lateral trochlear ridge, and tibial tubercle-trochlear groove distances. Values were compared between skeletally immature and mature groups. RESULTS: Sixty-eight children (22 girls, 46 boys, age: 14.0 ± 1.7 years) yielded 74 knees with patellar osteochondritis dissecans lesions, 14 (19%) of which were skeletally mature. The most common anatomic location was over the central patella [median ridge (34/74 - 46%) on the axial images and over the middle third (45/74 - 61%) on the sagittal images]. Overall, mean trochlear sulcus angle (high, 151 ± 11°), trochlear depth index (low, 2.8 ± 1.4 mm), and Insall-Salvati index (borderline, 1.3 ± 0.1) were abnormal for the entire sample. Skeletally mature knees were significantly more likely to have higher (more dysplastic) Dejour types when compared to skeletally immature knees (p < 0.01). Knees in the mature group, compared to immature, had significantly more abnormal mean lateral trochlear inclination (15 ± 8° vs. 19 ± 6°, p = 0.03) and patellar tendon-lateral trochlear ridge distance (5.55 ± 4.31 mm vs. 2.89 ± 4.69 mm, p = 0.04). Half of the knees had ≥ 4 abnormal features that predispose to patellofemoral maltracking; mature knees were significantly (p = 0.02) more likely to have a higher number of abnormal features (> 6 features, 7/14, 50.0%) versus immature knees (0-3 features, 33/60, 55.0%). CONCLUSION: In children with patellar osteochondritis dissecans, abnormal patellofemoral morphology and alignment indices were common in all patients and more severe in mature knees.

BACKGROUND: Becoming a first-level discipline in China means access to more educational resources. The development of medical humanities in China has been going on for more than 40 years, and some medical schools have set up master's and doctoral programs in medical humanities. The demand for medical humanities-related knowledge in China is also growing after COVID-19. However, medical humanities is only a second-level discipline and receives limited resources to meet the needs of society. This study aims to establish a system of indicators that can assess whether the medical humanities has a first-level discipline and provide a basis for its upgrading to a first-level. METHODS: A Delphi technique was used, with the panel of expert expressing their views in a series of two questionnaires. A coefficient of variation of less than 0.2 indicates expert agreement. RESULT: A total of 25 experts participated in this Delphi study. Consensus was reached on 11 first-grade indices and 48 s-grade indices. The authoritative coefficient(Cr) of the experts was 0.804, which indicates that the experts have a high level of reliability. CONCLUSION: This study provides a reliable foundation for the evaluation of medical humanities maturity.

This study, for the first time, has investigated the relationships between alterations of mangiferin contents in mango leaves at different maturity stages and their antibacterial properties. Leaves were classified into six different maturity stages based on their color: (1) young dark reddish brown, (2) young yellow, (3) young light green, (4) mature green, (5) old dark green, and (6) old yellow leaves. Ethanol extracts were then examined against Gram-positive and Gram-negative bacteria, applying broth dilution and agar well diffusion methods. In addition, we also measured the mangiferin contents in leaves at different stages for the purpose of evaluating how the changes in this phytochemistry value affects their activities against bacteria. The results showed that extracts from leaves at young ages had better antibacterial properties than those from old leaves, as evidenced by the lower minimum inhibitory concentrations and larger inhibitory zones. In addition, we also found that the contents of mangiferin were significantly decreased followed the maturation process. These results suggest that mango leaves at young stages, especially dark reddish brown and young yellow leaves, are preferable for application in bacterial infections and other therapies related to mangiferin's constituents.

To identify potential heavy minerals in the Mbiame floodplain in the Northwestern region of Cameroon, geochemical and petrographical data from alluvial deposits were utilized. The grains are mixed with inclusions of zircon and ilmenite, and the megacrysts come from felsic to intermediate igneous rocks like old basalt-andesite and magmatic gneiss. The Mbiame region lies between latitudes 6° 8' and 6° 24' N and longitudes 10° 39' and 10° 57' E, East of Mount Oku. One of the principal volcanic massifs on the Cameroon Volcanic Line (CVL), Mt. Oku is situated over a basement composed of migmatites, biotite diorites, and Pan-African granite-gneisses that date back to 600 Ma. The Mbiame Floodplain is located in the Pan African Fold Belt, which is made up of the Benin-Nigeria belt, which is a component of the Central African Fold Belt, and the Trans-Saharan belt. Between the Congo and West African cratons is a heavily eroded and deformed combination of Neoproterozoic plutonic and volcanic arcs and Paleoproterozoic micro-continents. The Central Cameroon Shear Zone serves as the primary lineament for the Pan African Orogeny, which is structurally separated into the Northwestern Cameroon Domain (NWC), the Adamawa-Yade Domain (AYD), and the Yaounde Domain (YD) in Cameroon. The field work was carried out during the dry season. During the field campaign, numerous bulk sediments were sampled from three (03) pits of vertical lithological succession along the left terrace of the Mbiame plain and twelve (12) were selected for laboratory analyses. The criteria for sample collection were based on texture (grain size) and color parameters. To identify the heavy minerals in the sediments and to understand the source of the sediments, the selected 12 bulk samples from the different layer of the pits were dried and sieved for heavy mineral determination. Heavy mineral separation was performed on about 2 g of the concentrate using bromoform liquid and the retained heavy minerals were mounted and observed under a petrographic microscope for optical properties identification. Twelve (12) bulk sediment samples weighing 100 g each were ground up in a mild steel pulverizer before being examined for major and trace elements. At Acme Analytical Laboratories Ltd., Vancouver, Canada, the geochemical study was carried out utilizing a combination of mass spectrometry (ICP-MS) for trace elements and inductively coupled plasma emission spectrometry (ICP-ES) for significant oxides. Lithium metaborate/tetraborate (LiBO2/Li2B4O7) flux was combined with 0.2 g of sample in crucibles, and the mixture was fused in a furnace. After the beads cooled, they were digested in ACS-grade nitric acid and then subjected to ICP-MS analysis. To calculate loss on ignition (LOI), a sample split at 1000 °C was ignited, and the weight loss was then measured. Data of this paper are further presented and discussed in Etutu et al. [1].

The current study aimed to describe the annual gametogenic phenology of the oyster Magallana bilineata (Röding, 1798) (=Crassostrea madrasensis), which is found on the west coast of Moheshkhali Island, Cox's Bazar, Bangladesh. Samples were drawn monthly from the intertidal region during low tide, from which 20 adult individuals were selected at random for biometry and histology. The mean condition index (CI), a ratio of tissue wet weight (g) to shell length (cm), varied from 0.58 ± 0.08 to 1.32 ± 0.36. Histology revealed two spawning cycles in the habitat of M. bilineata over the 12 months of the study. Gametogenesis initiated in December and May, and ripe animals principally occurred from July to September and February to May. In the habitat, M.bilineata exhibited two spawning peak periods: April to June and August to October. The undifferentiated stage as a preparatory step for the next spawning extended from November to February for the first spawning cycle and for a brief period in June for the next spawning cycle. The initiation of spawning in March could be associated with the high-level decline of salinity and increased temperature between February and March, in association with the annual rainfall start, whereas the next spawning cycle could be associated with a gradual increase in salinity. No spawning activity was reported from December to February, when the water temperature remained below 22 °C. Further study could be undertaken on the timing of spatfall of M.bilineata in the habitat to harvest spats for commercial farming of this promising species.

Hyperthermophilic composting, characterized by temperatures equal to or exceeding 75 °C, offers superior compost maturity and performance. Inoculation with thermophilic bacteria presents a viable approach to achieving hyperthermophilic composting. This study investigates the effects of inoculating thermophilic bacteria, isolated at different temperatures (50 °C, 60 °C, and 70 °C) into compost on maturity, gaseous emissions, and microbial community dynamics during co-composting. Results indicate that the thermophilic bacteria inoculation treatments exhibited peak temperature on Day 3, with the maximum temperature of 75 °C reached two days earlier than the control treatment. Furthermore, these treatments demonstrated increased bacterial richness and diversity, along with elevated relative abundances of Firmicutes and Proteobacteria. They also fostered mutualistic correlations among microbial species, enhancing network connectivity and complexity, thereby facilitating lignocellulose degradation. Specifically, inoculation with thermophilic bacteria at 60 °C increased the relative abundance of Thermobifida and unclassified-f-Thermomonosporaceae (Actinobacteriota), whereas Bacillus, a thermophilic bacterium, was enriched in the 70 °C inoculation treatment. Consequently, the thermophilic bacteria at 60 °C and 70 °C enhanced maturity by 36 %-50 % and reduced NH3 emissions by 1.08 %-27.50 % through the proliferation of thermophilic heterotrophic ammonia-oxidizing bacteria (Corynebacterium). Moreover, all inoculation treatments decreased CH4 emissions by 6 %-27 % through the enrichment of methanotrophic bacteria (Methylococcaceae) and reduced H2S, Me2S, and Me2SS emissions by 1 %-25 %, 47 %-63 %, and 15 %-53 %, respectively. However, the inoculation treatments led to increased N2O emissions through enhanced denitrification, as evidenced by the enrichment of Truepera and Pusillimonas. Overall, thermophilic bacteria inoculation promoted bacteria associated with compost maturity while attenuating the relationship between core bacteria and gaseous emissions during composting.

Whole-crop wheat (Triticum aestivum, WCW) has a high nutritional value and digestibility. No-tillage (NT) can reduces energy and labor inputs in the agricultural production process, thus decreasing production costs. There are many studies on planting techniques of WCW at present, few being on no-tillage planting. This study aimed to compare the effects of different tillage methods and maturity stages on the yield, nutritive value, and silage fermentation quality of WCW. The experiment included two tillage methods (NT; conventional tillage, CT), two maturity stages (flowering stage; milk stage), and three years (2016-2017; 2017-2018; 2018-2019). Years had a strong influence on the yield and nutritional composition of WCW. This was mainly related to the amount of rainfall, as it affects the seedling emergence rate of wheat. Although tillage methods showed no significant effects on the yield, plant height, and stem number per plant of WCW (P > 0.05), compared to CT, the dry matter (DM) and crude protein (CP) yields of NT decreased by 0.74 t/ha and 0.13 t/ha. Tillage methods showed no significant effects on the nutritive composition of WCW (P > 0.05). The WCW at the milk stage had greater DM (5.25 t/ha) and CP (0.60 t/ha) yields than that at the flowering stage (3.19 t/ha and 0.39 t/ha) (P< 0.05). The acid detergent fiber concentration of WCW decreased by 34.5% from the flowering to the milk stage, whereas water-soluble carbohydrates concentration increased by 50.6%. The CP concentration at the milk stage was lower than that at the flowering stage (P< 0.05). The lactic acid concentration of NT (17.1 g/kg DM) silage was lower than that of CT (26.6 g/kg DM) silage (P< 0.05). The WCW silage at the milk stage had a lower NH3-N concentration (125 g/kg TN) than that at the flowering stage (169 g/kg TN) (P< 0.05). Wheat sown by NT and CT was of similar yield and nutritional value, irrespective of harvest stages. WCW harvested at the milk stage had greater yield and better nutritional composition and silage fermentation quality than that at the flowering stage. Based upon the results of the membership function analysis, no-tillage sowing of wheat was feasible and harvesting at milk stage was recommended.

Hepatocyte nuclear factor 1α (HNF1α) is a transcription factor that is crucial for the regulation to maintain the function of pancreatic ß-cell, hepatic lipid metabolism, and other processes. Mature-onset diabetes of the young type 3 is a monogenic form of diabetes caused by HNF1α mutations. Although several mutation sites have been reported, the specific mechanisms remain unclear, such hot-spot mutation as the P291fsinsC mutation and the P112L mutation and so on. In preliminary studies, we discovered one MODY3 patient carrying a mutation at the c.493T>C locus of the HNF1α gene. In this study, we analyzed the pathogenic of the mutation sites by using the Mutation Surveyor software and constructed the eukaryotic expression plasmids of the wild-type and mutant type of HNF1α to detect variations in the expression levels and stability of HNF1α protein by using Western blot. The analyses of the Mutation Surveyor software showed that the c.493T>C site mutation may be pathogenic gene and the results of Western blot showed that both the amount and stability of HNF1α protein expressed by the mutation type plasmid were reduced significantly compared to those by the wild type plasmid (P<0.05). This study suggests that the c.493T>C (p.Trp165Arg) mutation dramatically impacts HNF1α expression, which might be responsible for the development of the disease and offers fresh perspectives for the following in-depth exploration of MODY3's molecular pathogenic process.

BACKGROUND: As part of the new Flanders Quality Model (FlaQuM) towards sustainable quality management systems, a co-creation roadmap with six primary drivers and 19 building blocks that guides healthcare organisations has been developed. Currently, no assessment tool is available to monitor hospitals' quality management systems implementation according to this co-creation roadmap. Therefore, we aimed to measure the maturity of the implementation of the FlaQuM co-creation roadmap in hospitals. METHODS: A three-phase approach in co-design with 19 hospitals started with defining the scope, followed by establishing content validity through a literature review, involvement of content experts (n=47), 20 focus groups with content experts (n=79) and a Delphi round with healthcare quality managers (n=19) to test the content validity index (CVI). Construct validity was assessed by confirmatory factor analyses and convergent validity by Spearman's ρ correlation coefficients. RESULTS: Based on 17 included existing maturity instruments and sub-components of content experts, two maturity tools were developed according to the implementation of the FlaQuM co-creation roadmap: 1) a maturity matrix with 52 sub-components and 2) a co-creation scan with 19 statements. The overall scale-CVI varied between 93.3% and 90.0% in terms of relevance and clarity, respectively. In a sample of 119 healthcare professionals, factor analyses revealed a 6-factor structure and 16 (84.2%) of the 19 hypothesis for testing convergent validity between both maturity tools were statistically significant. CONCLUSION: Measuring the implementation of the FlaQuM co-creation roadmap and monitoring its maturity over time should be feasible by using these comprehensive maturity tools in hospitals. Results of both tools should be able to describe the current state of hospitals' implementation of the co-creation roadmap as basis for strategic improvement plans and next steps.

PURPOSE: The Sanders Scoring System has revolutionized the way we assess the remaining growth potential of the skeleton. However, because it involves radiation exposure, it must be used with caution in children. The purpose of the study was to evaluate whether the Sanders skeletal maturity score (SMS) could be accurately determined using ultrasound (U). METHODS: We took radiographs (R) of the hand and performed U of the thumb and index finger in 115 patients between six and 19 years of age who were undergoing treatment for scoliosis or limb deformities. Paediatric orthopaedic surgeons, a paediatrician, and a paediatric radiologist were evaluated the blinded images. Those classified images are based on the SMS and the Thumb Ossification Composite Index (TOCI). RESULTS: Intrarater reliability was high for SMS and slightly weaker for TOCI, but still significant. Interrater reliability was clear for R and weaker for U in both staging systems. Ultimately, SMS 3 and 7 achieved the highest percentage of concordance (P) of 71.7% and 66.0%, respectively, when U was performed. Combining the clinically relevant groups of SMS 3&4 and SMS 7&8 also significantly increased peak scores (SMS 3 and 4 P = 76.7%; SMS 7 and 8 P = 79.7%). The probabilities of peak scores were significantly weaker when the TOCI score was examined. CONCLUSION: Our study shows that U can be used effectively especially to measure stages 3 and 4 and stages 7 and 8 of SMS. The U method is easy to use and therefore may offer advantages in clinical practice without the need for radiation exposure.

Grapevines are frequently subjected to heatwaves and limited water availability during ripening. These conditions can have consequences for the physiological health of the vines. Moreover, the situation is often exacerbated by intense solar radiation, resulting in reduced yield due to sunburn and a decline in quality. In light of these challenges, our study aimed to develop a fruit-zone cooling system designed to mitigate grape sunburn damage and improve the microclimate conditions within the vineyard. The system comprises a network of proximal sensors that collect microclimate data from the vineyard and an actuator that activates nebulizers when the temperature exceeds the threshold of 35°C. The research was conducted over two years (2022 and 2023) in Bologna (Italy) using potted Sangiovese and Montepulciano vines. These two vintages were characterized by high temperatures, with varying amounts of rainfall during the test period, significantly impacting the evaporative demand, which was notably higher in 2023. Starting from the veraison stage we compared three treatments: Irrigated control vines (WW); Control vines subjected to 50% water restriction during the month of August (WS); WS vines treated with nebulized water in the bunch area during the stress period (WS+FOG). The application of nebulized water effectively reduced the temperature of both the air around the clusters and the clusters themselves. As we expected, Montepulciano showed better single leaf assimilation rate and stomatal conductance under non-limiting water conditions than Sangiovese while their behavior was unaffected under water-scarce conditions. Importantly, for the first time, we demonstrated that nebulized water positively affected gas exchange in both grape varieties. In addition to this, the vines treated with the misting system exhibited higher productivity compared to WS vines without affecting technological maturity. In the 2023 vintage, the activation of the system prevented the ripening blockage that occurred in Montepulciano under water stress. Regarding the concentration of total anthocyanins, a significant increase in color was observed in WS+FOG treatment, suggesting a predominant role of microclimate on anthocyanin biosynthesis and reduction of oxidative phenomena. In conclusion, the fruit-zone cooling system proved to be an invaluable tool for mitigating the adverse effects of multiple summer stresses.

This research introduces a novel framework for enhancing soybean cultivation in North America by categorizing growing environments into distinct ecological and maturity-based zones. Using an integrated analysis of long-term climatic data and records of soybean varietal trials, this research generates a zonal environmental characterization which captures major components of the growing environment which affect the range of adaptation of soybean varieties. These findings have immediate applications for optimizing multi-environment soybean trials. This characterization allows breeders to assess the environmental representation of a multi-environmental trial of soybean varieties, and to strategize the distribution of testing and the placement of test sites accordingly. This application is demonstrated with a historical scenario of a soybean multi-environment trial, using two resource allocation models: one targeted towards improving the general adaptation of soybean varieties, which focuses on widely cultivated areas, and one targeted towards specific adaptation, which captures diverse environmental conditions. Ultimately, the study aims to improve the efficiency and impact of soybean breeding programs, leading to the development of cultivars resilient to variable and changing climates.
